Creating a Website Template

Is it possible in Drupal to build a website as a "template" and then copy it for each client so that it can be customized and themed for each unique implementation? In building out a microsite of specialized content that would then be marketed to target companies, would this be a better approach to take rather than building modules that can be reused. The focus of the microsite would really be on the content of the website rather than functionality.

What's a good source of information for learning how to template and copy a Drupal site?
